# Thermal Management for DNA Amplification

#### **Fast, Precise Temperature Cycling for PCR Thermal Cyclers**

Polymerase chain reaction (PCR) is a molecular biology technique used to amplify specific DNA sequences. PCR is performed in a **thermal cycler**, which repeatedly heats and cools DNA samples through precisely controlled temperature steps.

Modern PCR, qPCR and digital PCR instruments require **fast temperature ramp rates, accurate setpoint control and uniform temperature distribution** across the sample block. Thermoelectric coolers (TECs) provide bidirectional heating and cooling in a compact solid-state device, making them well suited for PCR thermal cycling.

## Why Precise Temperature Control Matters in PCR

A PCR cycle typically moves between temperature stages for DNA denaturation, primer annealing and DNA extension. Depending on the assay, these stages commonly occur around:

- **~94–98°C** for DNA denaturation
- **~50–65°C** for primer annealing
- **~68–72°C** for DNA extension

These temperature transitions are repeated many times during each PCR run. Fast and uniform thermal control can help reduce cycle time while maintaining consistent reaction conditions across the sample block.

For PCR instrument designers, key thermal requirements include:

- Fast heating and cooling ramp rates
- Precise, repeatable temperature control
- Low temperature gradients across sample wells
- Reliable operation through repeated thermal cycling
- Compact integration beneath PCR blocks or cartridges

### **Thermoelectric Cooling for PCR Thermal Cyclers**

Thermoelectric coolers use the Peltier effect to provide **both heating and cooling from the same solid-state device**. Reversing the electrical current reverses the direction of heat flow, allowing a PCR thermal cycler to move rapidly between programmed temperature setpoints.

However, repeated heating and cooling also causes the thermoelectric module and surrounding materials to expand and contract. These repeated thermal stresses can contribute to mechanical fatigue in conventional TECs and reduce operating life.
